Overview

LAH4 is a synthetic, amphipathic, alpha-helical peptide designed for potent antimicrobial activity and efficient nucleic acid transfection. Composed of 26 amino acids with the sequence KKALLALALHHLAHLALHLALALKKA-NH2, it features a leucine-rich hydrophobic face and a histidine-rich hydrophilic face. Its mechanism of action involves pH-dependent membrane interaction; at acidic pH, the protonation of histidine residues promotes membrane insertion and disruption, facilitating the endosomal escape of DNA or RNA cargoes. Primarily used as a research tool, LAH4 demonstrates high affinity for anionic lipids found in bacterial membranes and is frequently employed in laboratory settings to study cell penetration and non-viral gene delivery systems.
