LAIR-1 antibody-drug conjugates (ADCs) are a class of experimental therapeutic agents designed to target the leukocyte-associated immunoglobulin-like receptor 1 (LAIR-1, also known as CD305), an inhibitory immune receptor that is highly expressed on the surface of various hematologic malignancies, including acute myeloid leukemia (AML) and certain lymphomas. These ADCs typically consist of a LAIR-1-specific monoclonal antibody conjugated to a cytotoxic payload, such as the microtubule inhibitor monomethyl auristatin E (MMAE). The mechanism of action involves the binding of the ADC to LAIR-1 on cancer cells, followed by receptor-mediated endocytosis and lysosomal trafficking. Once inside the lysosome, the cytotoxic payload is released, leading to cell cycle arrest and apoptosis. Notable candidates include LA-057-MMAE, developed by Shanghai Junshi Biosciences in collaboration with Fudan University, and programs by NextCure, both of which have demonstrated potent anti-tumor activity and safety in preclinical models.
