LAIR-1 monoclonal antibody

LAIR-1 monoclonal antibody refers to a class of therapeutic agents, such as NextCure's NC525, that target the Leukocyte-associated immunoglobulin-like receptor 1 (LAIR-1). LAIR-1 is an inhibitory receptor expressed on hematopoietic cells and is frequently overexpressed in hematologic malignancies like acute myeloid leukemia (AML) and certain T-cell lymphomas, where it may provide a survival advantage to cancer cells. These antibodies can function as agonists to induce direct cell death in leukemic cells or be developed as antibody-drug conjugates (ADCs) to deliver cytotoxic payloads upon internalization. Preclinical data for LAIR-1 ADCs have shown significant anti-tumor activity and safety in humanized mouse models, demonstrating rapid internalization in tumor cells but not in healthy peripheral blood mononuclear cells (PBMCs). This supports the potential of LAIR-1 targeted therapies for treating LAIR-1-expressing hematological cancers.
