Lambda interferon adenovirus refers to a *gene therapy modality* using an adenoviral vector to express human interferon lambda (IFN-λ1 or IFN-λ2) in target tissues or cells. Lambda interferon (also called IFN-λ or type III interferon) is a cytokine with potent, epithelial-specific antiviral and immunomodulatory activity. The adenoviral vector delivers the IFN-λ gene to cells, leading to local production of interferon lambda protein. Mechanistically, IFN-λ binds to the interferon lambda receptor (IFNLR1/IL-10R2), activating the JAK/STAT pathway, upregulating antiviral and pro-apoptotic genes, and modulating immune responses with focus on epithelial barriers such as respiratory, hepatic, and gastrointestinal mucosa. In experimental cancer models, lambda interferon adenovirus induces tumor cell apoptosis and suppresses proliferation via STAT1/2/3 activation and the Bcl-2/Bax-caspase cascade[7]. In respiratory viral and cancer contexts, it enhances localized antiviral immunity while minimizing systemic inflammation.
