Overview
Lamofree ER is an extended-release formulation of lamotrigine, a phenyltriazine derivative primarily used as an anticonvulsant and mood stabilizer. Investigated by Taipei City Hospital for the treatment of ketamine use disorder (ketamine dependence), it functions by blocking voltage-gated sodium channels. This action stabilizes presynaptic membranes and inhibits the release of excitatory neurotransmitters, most notably glutamate. By modulating the glutamatergic system, Lamofree ER aims to counteract the neurobiological dysregulation and cravings associated with chronic ketamine use. The drug was evaluated in a Phase 2/3 randomized, double-blind, placebo-controlled trial over a 12-week period.
