LAMP5 CAR T-cells are an investigational chimeric antigen receptor (CAR) T-cell therapy targeting Lysosomal Associated Membrane Protein Family Member 5 (LAMP5). LAMP5 is a protein found to be aberrantly expressed on the surface of malignant plasma cells in multiple myeloma and in high-risk subtypes of acute myeloid leukemia (AML), such as KMT2A-rearranged AML, while showing minimal expression in normal hematopoietic cells. The therapy involves engineering T-cells with a CAR construct derived from a monoclonal antibody's single-chain variable fragment (scFv) specific to the extracellular domain of LAMP5. Preclinical studies have demonstrated that these CAR T-cells can specifically eliminate LAMP5-positive tumor cells with efficacy comparable to BCMA-targeted therapies in myeloma models. The therapy is being explored as a potential treatment for high-risk hematologic malignancies where LAMP5 serves as a mediator of clonogenicity and disease progression.
