Lampalizumab is an antigen-binding fragment (Fab) of a humanized monoclonal antibody that selectively binds to and inhibits complement factor D, a rate-limiting enzyme in the activation and amplification of the alternative complement pathway. It was developed as a potential treatment for geographic atrophy secondary to age-related macular degeneration. Lampalizumab was investigated in multiple phase 3 clinical trials (Spectri and Chroma), but these studies failed to meet their primary efficacy endpoints, leading to discontinuation of its development for this indication. The drug was originally developed by Tanox and later by Genentech/Roche[1][3][4][5][6].
