Langerin-targeted mRNA lipid nanoparticles (LNPs) are an experimental vaccine and therapeutic platform developed by Cutanos (based on research from the University of Vienna and Max Planck Institute). The system, known as the Langerhans Cell-Targeted Delivery System (LC-TDS), utilizes a glycomimetic small-molecule ligand to specifically target Langerin (CD207), a C-type lectin receptor expressed on skin-resident Langerhans cells. By selectively delivering antigen-encoding mRNA to these professional antigen-presenting cells in the epidermis, the platform enhances antigen translation and induces robust systemic CD8+ and CD4+ T-cell responses. This targeted approach allows for significant dose-sparing and potentially reduces systemic side effects by avoiding off-target accumulation in the liver, making it a promising candidate for both cancer immunotherapy and infectious disease vaccines.
