Overview

This is a four-drug oral combination therapy consisting of lansoprazole (a proton pump inhibitor), amoxycillin (a penicillin antibiotic), clarithromycin (a macrolide antibiotic), and tinidazole (a nitroimidazole antibiotic). It is primarily indicated for the eradication of Helicobacter pylori infection associated with peptic ulcer disease. Lansoprazole reduces gastric acid secretion by inhibiting the gastric proton pump, creating a less acidic environment that enhances the effectiveness of antibiotics against H. pylori. Amoxycillin disrupts bacterial cell wall synthesis, leading to bacterial death. Clarithromycin inhibits bacterial protein synthesis by binding to the 50S ribosomal subunit, thereby preventing growth and multiplication of bacteria. Tinidazole acts by damaging DNA and other critical biomolecules in susceptible microorganisms, particularly anaerobic bacteria and protozoa[5][6]. The combination targets H. pylori through multiple mechanisms to increase eradication rates and reduce resistance development.
