Lapatinib + capecitabine + vinorelbine is a combination regimen of three small molecule chemotherapeutic agents used primarily in the treatment of HER2-positive advanced or metastatic breast cancer. Lapatinib is an oral tyrosine kinase inhibitor that targets both the human epidermal growth factor receptor 2 (HER2) and epidermal growth factor receptor (EGFR), thereby inhibiting tumor cell proliferation. Capecitabine is an oral prodrug that is enzymatically converted to 5-fluorouracil (5-FU) in tumor tissue, where it inhibits DNA synthesis by blocking thymidylate synthase. Vinorelbine is a semi-synthetic vinca alkaloid that disrupts microtubule formation by binding to tubulin, leading to mitotic arrest and apoptosis in dividing cells[1][2][4][6]. This combination has been studied for its efficacy and tolerability as a later-line therapy for patients with HER2-positive metastatic breast cancer who have progressed on prior treatments including trastuzumab-based regimens.
