Overview

This regimen is a combination therapy consisting of lapatinib, cisplatin, and radiotherapy. **Lapatinib** is an oral small-molecule tyrosine kinase inhibitor that targets both epidermal growth factor receptor (EGFR, also known as ERBB1) and ERBB2 (HER2) receptors, thereby inhibiting downstream signaling pathways that promote cell proliferation and survival. **Cisplatin** is a platinum-based chemotherapeutic agent that induces DNA crosslinking, leading to apoptosis of rapidly dividing cancer cells. **Radiotherapy** uses ionizing radiation to damage the DNA of cancer cells, resulting in cell death. This combination has been clinically investigated—particularly in locally advanced, non–human papillomavirus (HPV) head and neck cancers—with the objective of enhancing anti-tumor efficacy by targeting distinct pathways and increasing overall cytotoxic effects. However, clinical trials conducted to date indicate that the addition of lapatinib to cisplatin and radiotherapy does not significantly improve progression-free or overall survival compared with chemoradiotherapy alone[1][2][3].
