Overview

Lapuleucel-T (APC8024, Neuvenge) is an investigational autologous cellular immunotherapy developed by Dendreon Corporation, designed to treat tumors that overexpress the HER-2/neu (ERBB2) receptor. The production process involves collecting a patient's peripheral blood mononuclear cells (PBMCs) via leukapheresis and culturing them ex vivo with BA7072, a recombinant fusion protein. This fusion protein consists of the intracellular and extracellular domains of HER-2/neu linked to granulocyte-macrophage colony-stimulating factor (GM-CSF), which serves to activate and direct antigen-presenting cells (APCs) against the target. Once re-infused into the patient, these activated cells are intended to stimulate a robust T-cell-mediated immune response against HER-2/neu-positive cancer cells. Although it reached Phase II clinical trials for indications such as metastatic breast, ovarian, and colorectal cancers, its development was discontinued.
