Large Multivalent Immunogen (LMI) is an experimental cancer vaccine platform consisting of tumor cell plasma membranes immobilized onto 5-μm amorphous silica microbeads. Developed by Lidak Pharmaceuticals (now Avanir Pharmaceuticals), LMI is designed to function as "pseudocells" that present a broad spectrum of tumor-associated antigens and MHC molecules to the immune system. The 5-μm size of the beads is specifically chosen to mimic the dimensions of a cell, which facilitates the direct activation of cytotoxic T lymphocytes (CTLs) or uptake by antigen-presenting cells for cross-priming. In clinical studies for metastatic melanoma, the LMI preparation utilized membranes from two specific melanoma cell lines (M-1 and M-2) and was administered to HLA-A2+ patients to stimulate tumor-specific immune responses without the need for traditional chemical adjuvants.
