Larotaxel + capecitabine is a combination of two anti-cancer drugs. Larotaxel is a taxane-class chemotherapy agent, structurally related to docetaxel and paclitaxel, and acts by stabilizing microtubules, thereby inhibiting cell division and triggering apoptosis. Capecitabine is an oral prodrug of 5-fluorouracil (5-FU) and is activated preferentially in tumor tissue by thymidine phosphorylase; it interferes with DNA synthesis via inhibition of thymidylate synthase. The combination aims to exploit synergy between the two mechanisms: taxane-mediated upregulation of thymidine phosphorylase may enhance activation of capecitabine within tumors, potentially increasing anti-tumor efficacy. This regimen has been investigated primarily for the treatment of solid tumors such as metastatic breast cancer and other advanced cancers resistant to first-line therapies[2][4].
