LAS-103 is a potent and selective endothelin B receptor (ETBR) antagonist developed using Lassogen's proprietary lasso peptide (lassotide) platform. Lasso peptides are engineered biologics with a unique knotted structure that confers high stability and protease resistance. LAS-103 targets ETBR, which is overexpressed on the endothelium of tumor vasculature in various "cold" tumors, including ovarian and triple-negative breast cancers. Activation of ETBR typically suppresses ICAM-1 expression, hindering immune cell extravasation; by blocking this receptor, LAS-103 increases immune cell infiltration into the tumor microenvironment. It is intended for use as a combination therapy to sensitize immunotherapy-resistant tumors to checkpoint inhibitors.
