LAS191954 is an investigational, orally administered small molecule inhibitor of phosphoinositide 3-kinase delta (PI3Kδ), developed by Almirall. It is primarily being evaluated for the treatment of autoantibody-mediated immunobullous skin diseases, most notably pemphigus vulgaris. By selectively targeting PI3Kδ, a kinase critical for immune cell signaling, LAS191954 aims to inhibit myeloid cell activation and the release of reactive oxygen species (ROS) mediated by immune complexes and C5a. Preclinical studies have demonstrated that the drug dose-dependently improves disease phenotypes in autoimmune blistering models and shows superiority over high-dose corticosteroids. It has entered Phase 1 clinical development to evaluate safety, tolerability, pharmacokinetics, and pharmacodynamics in healthy volunteers.
