Lassogen αvβ6 lassotide is an engineered lasso peptide (lassotide) developed by Lassogen to target the αvβ6 integrin, often as a dual inhibitor alongside αvβ8. These integrins are critical regulators of the tumor microenvironment, where they activate latent transforming growth factor beta (TGF-β), a potent immunosuppressive cytokine that drives resistance to immune checkpoint inhibitors. Lassotides are a unique class of ribosomally synthesized and post-translationally modified peptides (RiPPs) characterized by a threaded-ring or 'loop-over-ring' topology, which confers exceptional stability against proteolytic degradation and thermal stress. By selectively antagonizing αvβ6, this therapeutic aims to block TGF-β activation, thereby sensitizing 'cold' or immunotherapy-resistant tumors to checkpoint blockade. Preclinical studies of lead candidates, such as Lassotide 47 and Lassotide 19, have demonstrated robust tumor regression and growth inhibition in mouse models of triple-negative breast and ovarian cancers when administered in combination with anti-PD-1 therapy.
