Lassogen αvβ8 lassotide is an engineered lasso peptide (lassotide) designed to selectively inhibit the αvβ8 integrin receptor. Developed by Lassogen, this therapeutic candidate targets the αvβ8-mediated activation of latent TGFβ (Transforming Growth Factor beta) within the tumor microenvironment. By blocking this pathway, the drug aims to alleviate TGFβ-induced immunosuppression, thereby sensitizing "cold" or immunotherapy-resistant tumors to the immune system. It is primarily intended for use in combination with checkpoint inhibitors to treat a variety of challenging malignancies, including colon cancer, breast cancer, metastatic melanoma, and squamous cell tumors. Preclinical studies, particularly with candidates like lassotide 47, have demonstrated the ability to halt tumor growth and induce regression in mouse models of triple-negative breast and ovarian cancers.
