LAT8881 is a synthetic 16–amino acid cyclic peptide analogue of a C-terminal fragment of human growth hormone (sequence YLRIVQCRSVEGSCGF, including two disulphide-linked cysteines). It was originally developed for obesity but is now being investigated primarily for neuropathic pain and migraine. Unlike full-length growth hormone, LAT8881 does not act via the classical growth hormone receptor. Instead, it interacts with the Lanthionine synthetase C-like (LANCL) protein family, which are peptide-modifying enzymes involved in supporting cell survival under oxidative and chemical stress. Preclinical studies have shown that LAT8881 enhances cell viability during cytotoxic stress and has demonstrated efficacy in animal models of neuropathic pain as well as viral infection models by reducing inflammation and tissue damage. The drug has been administered orally and intravenously in clinical trials, showing an excellent safety profile with no serious adverse events reported[1][6][8].
