LAVA-051 is a humanized bispecific single-domain antibody (Gammabody) designed to engage and activate both Vγ9Vδ2 T cells and type 1 natural killer T (NKT) cells to target and kill CD1d-expressing tumor cells. The molecule consists of two single-domain antibodies linked by a short glycine-serine linker—one domain binds the Vδ2 chain of the Vγ9Vδ2 T cell receptor, while the other targets CD1d, a glycoprotein involved in antigen presentation on hematological malignancies such as chronic lymphocytic leukemia (CLL), multiple myeloma (MM), and acute myeloid leukemia (AML). Its mechanism leverages dual engagement for potent antitumor activity with a low risk of cytokine release syndrome. Developed using Lava Therapeutics' Gammabody platform, it was evaluated in phase 1/2a clinical trials for relapsed/refractory CLL, MM, and AML[3][6][7][8].
