LAVA-1207 is a humanized bispecific antibody designed to engage both the Vδ2 chain of Vγ9Vδ2-T cells (a potent subset of gamma-delta T cells) and prostate-specific membrane antigen (PSMA), which is highly expressed on prostate cancer cells. The molecule consists of two single-domain antibody fragments (VHHs) linked to a silenced human IgG1 Fc domain. By binding simultaneously to PSMA on tumor cells and the Vδ2-T cell receptor, LAVA-1207 redirects and activates Vγ9Vδ2-T cells to mediate targeted cytolytic activity against PSMA-expressing prostate cancer cells while sparing normal tissues. It is being developed primarily for metastatic castration-resistant prostate cancer (mCRPC). Preclinical studies have shown robust antitumor activity with a favorable safety profile, including low risk for high-grade cytokine release syndrome. Clinical trials are ongoing in monotherapy as well as in combination with low-dose interleukin-2 or pembrolizumab[1][3][5][6][7].
