LAVA-1266 is a potent bispecific T cell engager (bsTCE) developed by LAVA Therapeutics using its Gammabody platform. It is designed to target and destroy CD123-positive tumor cells by engaging Vγ9Vδ2 T cells, a subset of gamma-delta T cells with anti-tumor activity. The drug selectively lyses CD123+ tumor cells with high potency and aims for a wide therapeutic window, minimizing cytokine release compared to traditional CD3-based engagers. Preclinical studies have shown that LAVA-1266 induces strong tumor cell lysis and activates Vγ9Vδ2-T cells without co-stimulating immunosuppressive regulatory T cells. Its primary indications are acute myeloid leukemia (AML) and myelodysplastic syndromes (MDS), where it is currently being evaluated in Phase 1 clinical trials[1][3][4][5][7].
