Drug intelligence / Profile preview
LAVR-289 is a novel, orally bioavailable acyclic nucleoside phosphonate (ANP) prodrug designed as a broad-spectrum antiviral agent targeting double-stranded DNA (dsDNA) viruses. Derived from the structure of cidofovir, LAVR-289 exhibits significantly lower toxicity and a high selectivity index. It acts by inhibiting viral DNA polymerase, functioning as a fraudulent building block during viral DNA synthesis to block replication firing and prevent the transition from early to late replication phases. LAVR-289 has demonstrated potent nanomolar in vitro and ex vivo efficacy against a wide range of dsDNA viruses, including human cytomegalovirus (HCMV), herpes simplex virus 1 and 2 (HSV-1/2), varicella-zoster virus (VZV), adenoviruses, poxviruses (such as monkeypox virus), and African swine fever virus (ASFV). It is being developed for the prevention and treatment of congenital HCMV infections, as well as opportunistic viral infections in immunocompromised patients.
