LB-100 + doxorubicin is an investigational combination therapy for cancer, specifically being studied in advanced soft tissue sarcomas. LB-100 is a small molecule inhibitor of protein phosphatase 2A (PP2A) and also inhibits PPP5C, both of which are serine/threonine phosphatases involved in cell signaling and regulation. By inhibiting these targets, LB-100 increases stress signals in tumor cells and can enhance the effectiveness of cytotoxic agents like doxorubicin. Doxorubicin is a well-established anthracycline chemotherapeutic agent that intercalates DNA and inhibits topoisomerase II, leading to DNA damage and apoptosis in rapidly dividing cells. Preclinical studies have shown that combining LB-100 with doxorubicin results in synergistic anti-tumor effects without significant additional toxicity compared to doxorubicin alone. The combination is currently under clinical investigation as a first-line treatment for advanced soft tissue sarcomas[1][2][3][4][5][6].
