Overview

LB-2.1 is a novel small molecule dopamine D2 and D3 receptor antagonist being developed by LB Pharmaceuticals. It belongs to the benzamide class of antipsychotics and is designed as a second-generation follow-on to amisulpride. Unlike amisulpride, which has poor blood-brain barrier permeability, LB-2.1 (and its sibling LB-102) are engineered to have improved lipophilicity and central nervous system (CNS) penetration. The drug is primarily intended for the treatment of schizophrenia, aiming to provide the efficacy of amisulpride (a widely used antipsychotic in Europe and Asia) with a more favorable pharmacokinetic profile for the U.S. market. While LB-102 is the lead candidate in clinical trials, LB-2.1 represents a distinct chemical entity within the company's portfolio of benzamide derivatives.
