LB-ECL04 is a recombinant endolysin-based antibacterial therapy being developed by LysinBio for the treatment of multidrug-resistant Gram-negative bacterial (MDR-GNB) infections, including pneumonia, bacteremia, and urinary tract infections. The therapeutic candidate is engineered using LysinBio's customized endolysin selection technology (CMic Tech) and consists of a bacteriophage-derived endolysin fused with papiliocin (PPLC), an antimicrobial peptide. This fusion design overcomes the outer membrane barrier of Gram-negative pathogens, allowing the endolysin to access and hydrolyze the peptidoglycan layer of the bacterial cell wall, leading to rapid osmotic lysis and cell death. LB-ECL04 has demonstrated potent antibacterial activity against key MDR pathogens such as Acinetobacter baumannii, Pseudomonas aeruginosa, Escherichia coli, and Klebsiella pneumoniae in preclinical studies.
