Overview

LB1410 + LB4330 is an investigational combination immunotherapy regimen under clinical development for advanced or metastatic solid tumors. The combination consists of two novel agents: - **LB1410**: A bispecific antibody targeting both Programmed cell death protein 1 (PD-1) and T-cell immunoglobulin and mucin domain-containing protein 3 (TIM3), designed to overcome resistance to anti-PD-1 therapies by simultaneously blocking two immune checkpoints. - **LB4330**: A bi-functional fusion protein that combines high-affinity binding to Claudin18.2 (CLDN18.2) with interleukin 10 (IL-10) activity. It is engineered to enrich in CLDN18.2-positive tumor microenvironments, activate CD8+ T cells, promote their proliferation and interferon secretion, and induce immune memory. The combination aims to enhance anti-tumor immunity by both releasing checkpoint inhibition (via PD-1/TIM3 blockade) and stimulating effector T cell responses within the tumor microenvironment (via CLDN18.2 targeting and IL-10-mediated activation). Clinical trials are ongoing in China for a range of advanced/metastatic solid tumors including pancreatic ductal adenocarcinoma, cholangiocarcinoma, colorectal cancer, ovarian cancer, non-small cell lung cancer, gastric/gastroesophageal junction adenocarcinoma, esophageal squamous cell carcinoma, hepatocellular carcinoma, renal cell carcinoma, cervical squamous cell carcinoma, and endometrial carcinoma[5][6][8].
