LB1410 + lenvatinib + LB4330 is a multi-pronged combination therapy developed by L&L Biopharma to treat advanced cancers that have become resistant to anti-PD-1/PD-L1 therapies. The regimen consists of **LB1410**, a recombinant humanized bispecific antibody that simultaneously blocks the Programmed Cell Death Protein 1 (PD-1) and T-cell Immunoglobulin and Mucin-domain containing-3 (TIM-3) pathways; **lenvatinib**, a small-molecule tyrosine kinase inhibitor (TKI) targeting Vascular Endothelial Growth Factor Receptors (VEGFR 1-3) to inhibit angiogenesis; and **LB4330**, a long-acting Interleukin-10 (IL-10) cytokine fusion protein designed to modulate the tumor microenvironment. This combination is being evaluated in Phase 2/3 trials for anti-PD-1 resistant cervical cancer and renal cell carcinoma, aiming to overcome immune evasion and reactivate anti-tumor immunity through dual checkpoint blockade, anti-angiogenesis, and cytokine-mediated immunomodulation.
