LB2402 is an autologous, bispecific chimeric antigen receptor (CAR) T-cell therapy developed by Legend Biotech for the treatment of relapsed or refractory multiple myeloma. The therapy is engineered to simultaneously target two antigens: CD19, a well-validated B-cell marker, and GPRC5D (G protein-coupled receptor class C group 5 member D), which is highly expressed on malignant plasma cells. By dual-targeting these antigens, LB2402 aims to mitigate the risk of antigen escape—a common mechanism of resistance in single-target CAR-T therapies—and provide more durable clinical responses. The program is currently in the clinical follow-up stage following investigator-initiated studies.
