LB4330 is a novel, bi-functional fusion protein designed to target Claudin18.2 (CLDN18.2) and modulate the tumor microenvironment by engaging CD8+ T cells and inhibiting interleukin 10 (IL-10). It acts as both a CLDN18.2 inhibitor and a CD8 T-cell agonist, with additional IL-10 modulatory activity. The drug demonstrates high affinity for human CLDN18.2 (14 pM) and is intended to activate tumor antigen-associated CD8+ T cells within the tumor microenvironment, potentially enhancing anti-tumor immune responses in solid tumors with low immunogenicity such as pancreatic ductal adenocarcinoma or colorectal cancer. Developed by L&L Biopharma, it is being evaluated alone or in combination with other immunotherapeutic agents—including PD1/PD-L1 monoclonal antibodies—for advanced solid tumors in ongoing clinical trials[1][4][5][6].
