Overview

LB54640 is an orally bioavailable, selective small-molecule agonist of the melanocortin 4 receptor (MC4R), currently under development by LG Chem. The MC4R is a G protein-coupled receptor primarily expressed in the hypothalamus, where it plays a critical role in the regulation of energy homeostasis, appetite, and body weight. Dysregulation of the MC4R pathway is a known cause of severe, early-onset obesity. LB54640 is designed to bypass upstream defects in the leptin-melanocortin signaling pathway, such as those seen in Bardet-Biedl syndrome (BBS) or deficiencies in pro-opiomelanocortin (POMC) and the leptin receptor (LEPR). Unlike existing peptide-based MC4R agonists that require daily subcutaneous injections, LB54640 offers the potential for oral administration, which may significantly improve patient adherence and quality of life. It is currently being evaluated in Phase 2 clinical trials for both rare genetic forms of obesity and general obesity.
