Overview

LB80317 is the *active metabolite* of the antiviral prodrug LB80380, developed for the treatment of chronic hepatitis B. It is a nucleotide analogue of guanosine monophosphate (GMP) that acts by inhibiting viral replication after phosphorylation and incorporation into viral DNA. LB80317 is generated in vivo by oxidative metabolism of the parent drug LB80331, which itself is released from the oral dipivoxil ester prodrug LB80380. LB80317 has a long plasma half-life at steady state (45–62 hours), leading to sustained exposure and supporting once-daily oral dosing. Its antiviral activity is mediated by inhibition of hepatitis B virus (HBV) polymerase, blocking viral DNA synthesis, and thus suppressing HBV replication. About 40% of LB80317 in mouse liver is found in the phosphorylated (active) form. The compound is primarily excreted via urine[1][2][4][7].
