Overview

LBL-033 is a bispecific T-cell engager (BiTE) antibody that targets both MUC16 (mucin 16) and CD3. By binding in a 2:1 ratio to MUC16 and CD3, it enhances aggregation in MUC16-overexpressing tumor cells while reducing non-specific activation of CD3, which may improve efficacy and minimize systemic toxicity such as cytokine release syndrome. The drug is designed to specifically target and kill MUC16-positive cancer cells by promoting immune cytokine secretion and modifying the tumor microenvironment to favor anti-tumor immunity. MUC16 is highly expressed in several malignant tumors including ovarian epithelial cancer, fallopian tube cancer, primary peritoneal cancer, endometrial cancer, pancreatic cancer, non-small cell lung cancer, and gastric cancer. LBL-033 was developed by Leads Biolabs and is currently being evaluated in phase I/II clinical trials for advanced solid tumors including ovarian cancer[1][2][5][6].
