Overview

LBL-043 is a **novel bispecific antibody** developed by Nanjing Leads Biolabs, designed to target both **Leukocyte immunoglobulin-like receptor B4 (LILRB4)** on tumor cells and **CD3** on T cells[1][2][4][5][9]. Its mechanism involves simultaneous binding to **LILRB4 (expressed on leukemia and multiple myeloma cells)** and **CD3 (on T cells)**, thus redirecting T cells to selectively kill LILRB4-positive tumor cells through T cell engagement and activation. LBL-043 was developed using the proprietary LeadsBody technology in a 2:1 format: two VHH domains (high-affinity for LILRB4) and one scFv domain (low-affinity for CD3). Preclinical data demonstrate potent anti-tumor activity in models of multiple myeloma and acute myeloid leukemia (especially M4/M5 subtypes), with a favorable safety profile in animal models[2][5].
