LBL-051-S3 is a trispecific T-cell engager (TCE) antibody developed by Oblenio Bio, a joint venture between Aditum Bio and Leads Biolabs. Built using the LeadsBody Platform, the molecule simultaneously targets CD3 on T cells, CD19 on B cells, and BCMA on plasma cells. This trispecific approach is designed to achieve a comprehensive depletion of the B-cell lineage, including long-lived plasma cells, which are often missed by CD19-targeted therapies alone. The drug is intended for the treatment of refractory autoimmune diseases, aiming to provide a durable immune system reset. Preclinical studies in non-human primates have shown that LBL-051-S3 effectively depletes target cells with a favorable safety profile, characterized by minimal cytokine release. While LBL-051-S3 is a specific preclinical construct variant, the broader LBL-051 program has progressed into Phase 1 clinical trials.
