LBL-058 is a novel first-in-class T cell engager conjugate (TCE-ADC) being developed by Nanjing Leads Biolabs. It targets Delta-like ligand 3 (DLL3), a Notch pathway ligand highly expressed in small cell lung cancer (SCLC) and other neuroendocrine tumors, with minimal expression in normal cells. LBL-058 uses a bispecific antibody that bridges CD3 on T cells and DLL3 on tumor cells, activating T cells to attack DLL3-positive cancer cells while simultaneously delivering a cytotoxic topoisomerase I inhibitor payload directly to tumor cells, thus combining the mechanisms of TCE and ADC for potent, durable anti-tumor activity. It is in preclinical development for oncology indications, especially SCLC and neuroendocrine carcinoma.
