LBx-PA01 is an investigational CRISPR-Cas3–enhanced bacteriophage antimicrobial being developed by Locus Biosciences as a precision therapy targeting Pseudomonas aeruginosa, including multidrug-resistant strains that are a serious hospital-acquired infection threat. It consists of engineered bacteriophages programmed with guide RNAs to direct the Cas3 nuclease to P. aeruginosa DNA, causing extensive, irreversible genome degradation in addition to the natural lytic activity of the phages, thereby selectively killing the pathogen while sparing non-target bacteria.[5][6] The program was supported by a partnership with IDbyDNA to develop a metagenomic companion diagnostic (based on the Explify platform) to identify P. aeruginosa–infected patients most likely to benefit from LBx-PA01 in clinical trials.[5][6][10][13]
