LC-101 is a pegylated liposomal formulation of doxorubicin hydrochloride, developed by Ayana Pharma as a generic equivalent to the reference product Doxil. As an anthracycline chemotherapy agent, doxorubicin functions as a topoisomerase II inhibitor and DNA intercalator, which inhibits macromolecular biosynthesis and leads to cell death. The pegylated liposomal delivery system significantly alters the drug's pharmacokinetic profile, allowing for prolonged circulation and increased accumulation in tumor tissues via the enhanced permeability and retention (EPR) effect, while simultaneously reducing the risk of cardiotoxicity compared to conventional doxorubicin. LC-101 received FDA approval in 2021 and is indicated for the treatment of ovarian cancer that has progressed or recurred after platinum-based chemotherapy, AIDS-related Kaposi's sarcoma, and multiple myeloma.
