LC007 is a mouse monoclonal antibody, and the precursor to the humanized antibody M4-3-ML2, targeting the membrane-proximal D3 domain of Melanoma-associated Chondroitin Sulfate Proteoglycan (MCSP), also known as Chondroitin Sulfate Proteoglycan 4 (CSPG4). Developed by Roche (specifically Roche Glycart), the antibody was engineered using GlycoMab technology to enhance its affinity for FcγRIIIa, thereby significantly increasing its potency in inducing antibody-dependent cellular cytotoxicity (ADCC). MCSP is a transmembrane proteoglycan highly expressed in 60-80% of melanomas, as well as in glioblastoma, various sarcomas, and on the pericytes of tumor neovasculature, while maintaining low expression in normal tissues. Preclinical studies demonstrated that glycoengineered versions of LC007 and M4-3-ML2 provide superior anti-tumor efficacy and survival benefits in disseminated melanoma models compared to non-glycoengineered counterparts.
