Drug intelligence / Profile preview
LC16m8Δ-AAV5 is an experimental heterologous prime-boost viral-vectored vaccine regimen designed for the prevention of malaria and the blocking of parasite transmission [1003]. Developed by researchers at Kanazawa University, Jichi Medical University, and Hokkaido University, the regimen utilizes a priming dose of the highly attenuated, replication-competent vaccinia virus strain LC16m8Δ (m8Δ) followed by a booster dose of an adeno-associated virus serotype 5 (AAV5) vector. Both vectors are genetically engineered to express a fusion protein containing the pre-erythrocytic stage antigen Plasmodium falciparum circumsporozoite protein (PfCSP) and the sexual-stage transmission-blocking antigen Pfs25. In preclinical studies, this prime-boost strategy has demonstrated robust immunogenicity, eliciting high titers of functional antibodies, providing sterile protection against sporozoite challenge, and showing potent transmission-blocking activity.
