LC28-0126 (also known as NecroX-7 or MIT-001) is a synthetic small molecule classified as an indole-thiazole derivative. It acts primarily as a reactive oxygen species (ROS) scavenger and necrosis inhibitor. The drug exhibits multiple mechanisms of action including inhibition of NF-kappa B signaling, NFATC transcription factors, proto-oncogene protein c-Fos, and HMGB1 (high-mobility group box 1). These actions contribute to its anti-inflammatory and cytoprotective effects. LC28-0126 is being developed for the treatment of conditions associated with oxidative stress and cell death such as acute myocardial infarction (reperfusion injury), oral mucositis, graft-versus-host disease (GVHD), hepatic fibrosis, pulmonary fibrosis, nephrotoxicity induced by FK506/tacrolimus in kidney cells, chronic colitis via attenuation of pro-inflammatory cytokines like TNF-alpha and IL1-beta expression. The drug has demonstrated neuroprotective effects in preclinical models for neurological disorders[1][2][3][4][5][7][8].
