Drug intelligence / Profile preview
LC30 is an orally bioavailable, non-kinase small molecule inhibitor developed by Anticancer Bioscience for the synthetic lethal targeting of MYC-expressing tumors. It is designed to exploit the genetic vulnerabilities of cancers overexpressing the MYC oncogene. In dividing cells, LC30 disrupts the localization of the chromosomal passenger protein complex (CPPC)—which consists of Aurora B kinase (AURKB), inner centromere protein (INCENP), survivin, and borealin—without inhibiting its catalytic activity. This disruption leads to mitotic defects, including multipolar spindles, micronuclei, polyploidy, mitotic arrest, and apoptosis. In resting cells, LC30 disrupts the Golgi structure, which may relate to MYC-mediated control of centrosomal gene expression, and exhibits anti-angiogenic effects by inhibiting endothelial cell migration and tubule formation. LC30 has demonstrated preclinical efficacy in various liquid and solid tumor models, including B- and T-cell malignancies, lung, colon, gastric, breast, and liver cancers.
