LCA-0321 is a lysosome-targeted chimera (LYTAC) degrader developed by Lycia Therapeutics. It is designed to specifically bind and rapidly eliminate disease-causing anti-thyroid-stimulating hormone receptor (TSHR) autoantibodies, which are the underlying cause of Graves' disease and its extrathyroidal manifestations such as thyroid eye disease (TED). By targeting these pathogenic autoantibodies for lysosomal degradation, LCA-0321 aims to restore normal thyroid function without causing general immunosuppression. Preclinical data have shown that it can selectively deplete patient-derived TSHR autoantibodies in both mice and human cells[3][5][6][8].
