Overview

LCA-0391 is a LYTAC (Lysosome Targeting Chimera) degrader being developed by Lycia Therapeutics for the treatment of MuSK-positive (MuSK+) myasthenia gravis. It is designed to selectively target and degrade anti-muscle-specific kinase (MuSK) autoantibodies, which are pathogenic drivers of this rare autoimmune neuromuscular disease. By utilizing the LYTAC platform, LCA-0391 recruits the cation-independent mannose-6-phosphate receptor (CI-M6PR) to shuttle the extracellular autoantibodies to the lysosome for degradation, thereby reducing the circulating levels of the disease-causing antibodies. As of 2024, the program has been designated as a development candidate and is in preclinical development.
