Overview

LCAR-LC12 is an autologous chimeric antigen receptor (CAR) T-cell therapy targeting C-type lectin-like molecule-1 (CLL1; also known as CLEC12A), developed by Legend Biotech. It is being investigated for the treatment of relapsed or refractory acute myeloid leukemia (AML). CLL1 is a type II transmembrane glycoprotein that is highly expressed on the surface of AML blasts and leukemic stem cells (LSCs) but is absent on normal hematopoietic stem cells (HSCs), making it an ideal target for reducing the risk of long-term myelosuppression. LCAR-LC12 consists of T cells engineered to express a CAR that recognizes CLL1, leading to the targeted destruction of malignant cells. Clinical trials, including those conducted at Anhui Provincial Hospital, have evaluated its safety and preliminary efficacy in patients with advanced AML, often in the context of bridging to allogeneic hematopoietic stem cell transplantation.
