LCAR-T2C is an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target the CD4 antigen. Developed by The First Affiliated Hospital with Nanjing Medical University, often in association with the LCAR platform from Legend Biotech, this therapy is specifically aimed at treating relapsed or refractory CD4-positive T-cell malignancies. These include T-cell non-Hodgkin lymphoma (T-NHL) and T-cell acute lymphoblastic leukemia (T-ALL). The treatment involves the ex vivo genetic modification of a patient's own T cells to express a CAR that recognizes CD4, enabling the immune system to selectively identify and eliminate malignant CD4+ cells. Due to the expression of CD4 on healthy helper T cells, the therapy carries a risk of profound T-cell aplasia and subsequent opportunistic infections, which are monitored during clinical evaluation. It is currently being assessed in Phase I clinical trials to determine its safety, tolerability, and preliminary efficacy.
