LCB02A is a Claudin 18.2 (CLDN18.2)-directed antibody-drug conjugate (ADC) being developed by LigaChem Biosciences (formerly LegoChem Biosciences). It consists of a humanized IgG1 monoclonal antibody (PR301839), licensed from Harbor BioMed, that specifically targets CLDN18.2, an isoform frequently overexpressed in various solid tumors such as gastric and pancreatic cancers. The antibody is conjugated to a cytotoxic topoisomerase 1 inhibitor (TOP1i) payload using LigaChem's proprietary ConjuAll site-specific conjugation technology, with a drug-to-antibody ratio (DAR) of 4. LCB02A is designed to selectively deliver the potent payload to tumor cells, minimizing off-target toxicity. It is currently in Phase 1/2 clinical development for the treatment of CLDN18.2-positive advanced solid tumors.
