LCB14-2516 is an antibody-drug conjugate (ADC) targeting B-cell maturation antigen (BCMA), developed for the treatment of multiple myeloma. It consists of an Fc-silenced IgG1 anti-BCMA monoclonal antibody conjugated to a highly potent pyrrolobenzodiazepine (PBD) DNA cross-linking payload using the ConjuAll platform. The ConjuAll technology utilizes a second-generation β-glucuronide (LBG) linker to ensure site-specific conjugation and stability. Preclinical studies indicate that LCB14-2516 exhibits superior efficacy compared to belantamab mafodotin biosimilars in various multiple myeloma models, with the potential for a reduced risk of ocular toxicity (keratopathy) often associated with earlier BCMA-targeted ADCs.
