Overview

LCB14-2524 is a next-generation antibody-drug conjugate (ADC) targeting B-cell maturation antigen (BCMA), currently in preclinical development for the treatment of multiple myeloma. Developed by LigaChem Biosciences (formerly LegoChem Biosciences) and AntibodyChem Biosciences, the molecule utilizes the proprietary ConjuAll platform, which features a second-generation β-glucuronide (LBG) linker to deliver a monomethyl auristatin F (MMAF) microtubule inhibitor payload. LCB14-2524 is designed with an ADCC-enhanced IgG1 antibody and maintains a homogenous drug-antibody ratio (DAR) of 2. Preclinical studies indicate that LCB14-2524 demonstrates superior efficacy compared to belantamab mafodotin biosimilars in various multiple myeloma models, with a design intended to improve the therapeutic index and reduce the risk of ocular toxicities, such as keratopathy, commonly associated with earlier BCMA-targeted ADCs.
