Overview

LCB22A is an antibody-drug conjugate (ADC) developed by LegoChem Biosciences in collaboration with GO Therapeutics. It is composed of the monoclonal antibody GO-013, which selectively targets cancer-specific Tn-glycosylated O-glycoprotein epitopes, conjugated to the microtubule-disrupting payload monomethyl auristatin E (MMAE). The conjugation utilizes LegoChem's proprietary ConjuAll technology, which features a site-specific, beta-glucuronidase-cleavable linker designed to ensure plasma stability and facilitate tumor-selective payload release. LCB22A is intended for the treatment of various solid tumors, including breast, lung, and colorectal cancers, where aberrant glycosylation results in the expression of tumor-associated carbohydrate antigens (TACAs). Preclinical studies have demonstrated target-specific cytotoxicity and tumor elimination in patient-derived xenograft (PDX) models, as well as a favorable safety profile in non-human primate toxicity studies.
